Create WDM device for the required HW inputs/outputs, because Windows/Skype do not support ASIO.
Browse manual for "WDM", "settings dialog", maybe you also need "loopback".
I was also searching for a solution! I used a WDM device to resolve this problem. It's working all very well. I needed it very much because I had a team-building meeting using a virtual conference platform. I don't know if you heard about online team-building games, but believe me that this is one of the greatest things that I discovered. I had a lot of fun last week because I participated in a few such events. In this way, I discovered a few new people that taught me some interesting IT things. Also, I think that these games will develop some of our communication skills.
Nice to hear, enjoy
Glad to hear you resolved it.
For anyone looking for more information in the future, I made a short video explaining what the channels correlate to on your audio interface and how to set them up for live audio applications: https://youtu.be/gOw7nu5O7AI

Thanks - very nice and helpful video. It's great to see increasing number of RME related instructional and very practical videos becoming available.
I also have a video discussing setting up TotalMix with video conferencing apps, and merging in both computer and external sources: https://youtu.be/Kj8eZ_fbOFw
In my case, I am using a Digiface Dante, but the principals are the same for all RME interfaces.
Synthax Audio UK also release a video in April 2020: https://www.youtube.com/watch?v=fCivF5Ql9CY
