When using "SUB" layout, the adding a route from a Input to this sub mix Output with the Matrix (or within another TMFx Mixer window), the new route isn't visible in the SUB layout, it becomes visible only when clicking SUB layout again.

Correct. As you can't change any routing outside the currently shown channels this is to be expected. A constant sync is not planned for that view. The sub view is there to work with the final mix, not to build the mix.
