Windows doesn't support ASIO, therefore you need to define WDM devices for Windows.

In short: assumed your active monitors are connected to HW output AN 1/2 ...
Once you configured WDM devices in the RME driver settings, then you find the speaker device in the Windows Sound Settings. Configure it as Windows Default sound device.
In TotalMix FX use the assign button to assign HW output AN 1/2 to "Main out".
By this you move this output to the control room to be able to use control room features like mute, dim, etc ...
To get a basic routing to Main out you can use predefined routings in TM FX:
- Reset Mix -> Total Reset
- Reset Mix -> Straight Playback (this is like "DAW mode")

- Reset Mix -> Straight Playback with all to Main Out
